In the course of biosynthesis of cobamides and their related compounds, a variety of intermediates can be found in addition to the final products. For the isolation, identification, and characterization of these substances, column chromatography, paper electrophoresis and physicochemical procedures such as ultraviolet and visible spectrophotometry have been employed. Sometimes, microbiological method may be used soly or in combination with bioautography. However, the microbiological method is not always adequate since some of natural or unnatural cobamides or also the intermediates of the biosynthesis exert rather an inhibitory action than no action on the growth of vitamin B12-requiring microorganisms. The authors are employing paper chromatography as well as paper electrophoresis in which the separated zones are detected or identified with the original color of vitamin B12. Because of a great interest in, and few reports (1, 2) on, the thin layer chromato graphy (TLC) of vitamin B12, fundamental investigations were made with special reference to the solvents as well as the adsorbents for the vitamin.
